Afghans extends deadline on private security ban

KABUL, Afghanistan (AP) ? The Afghan government is giving companies extensions ranging from a few weeks to 90 days to change from private security guards to a government-run force, NATO officials and security company managers said Sunday.

The reprieve comes just three days before the March 21 deadline that the Afghan government had set for the majority of companies to start using government-provided security.

Private development companies have said the move is threatening billions in U.S. aid to the country because companies would delay projects or leave altogether because they didn't feel safe using strictly local security.

President Hamid Karzai has railed for years against the large number of guns-for-hire in Afghanistan, saying private security companies skirt the law and risk becoming militias. He ordered them abolished in 2009 and eventually set the March deadline for all firms except those guarding military or diplomatic facilities to take on government guards.

But the process has been chaotic and has been weighed down by lengthy contract negotiations, making it appear unlikely in recent weeks that the Afghan Public Protection Force would be ready to take over for some 11,000 private guards by the deadline.

People involved in the handover said Sunday that companies that are still far from signing contracts are being allowed to continue to use private guards for a limited period of time, most ranging from 30 days to 90 days. Managers of private security companies, who spoke anonymously to avoid endangering contracts, said they had been called into a meeting with Afghan officials to explain the process for obtaining the interim permits.

Noorkhan Haidari, the business manager for the APPF, said the permit process was still under way and declined to comment further. Other APPF officials did not respond to calls seeking comment.

A number of deals have already been signed. So far, the APPF has signed 16 contracts with companies to provide security and licensed 14 "Risk Management Companies," according to a NATO official. The Risk Management Companies will essentially act as go-betweens for companies and the government agency in order to help manage the guards, payments and help hold the Afghan guards to an international standard.

The official spoke anonymously to discuss the inner workings of an Afghan government program.

But Afghan officials have said that there are about 75 companies they need to sign contracts with in order to complete the switchover and there were worries that holding to the March 21 deadline would create security gaps.

If the changeover doesn't happen smoothly, a raft of international aid projects could be in danger. Insurgents regularly attack development projects here, so private development companies that implement most of the U.S. aid agency's programs employ private guards to protect compounds, serve as armed escorts and guard construction sites.

Most companies working on development projects are being issued temporary 30-day permits for their private security guards, while companies working on convoy contracts are being given 90 days, the NATO official said. The development companies have a shorter extension because their projects tend to be easier to guard and many are already close to having a contract negotiated, the official said.

One element to the delay may have been that international military advisers who are helping the Afghan officials were blocked from going to the APPF offices following the shooting deaths last month of two Interior Ministry advisers by an Afghan driver. The APPF advisers are back at their offices now, though with some increased security, according to the NATO official.

Utah's Hatch battles Tea Party challengers in Republican caucus

SALT LAKE CITY, Utah (Reuters) - Veteran U.S. Senator Orrin Hatch faced what may prove to be the toughest re-election bid of his 36-year tenure, as he battled a challenge on Thursday by two younger Tea Party candidates in Utah's Republican caucus amid high turnout.

Heavily Republican Utah last elected a Democrat to the U.S. Senate more than four decades ago, so the victor in the state's Republican party contest is usually considered the presumptive winner of the general election in November.

For a senior stalwart of the Republican mainstream like Hatch, the ranking Republican on the tax-writing Senate Finance Committee, an incumbent seat in the U.S. Senate has long been seen as his to lose. He was first elected in 1976, turns 78 next week, and says this next term will be his last.

"We're starting to get numbers in and what we're seeing in the reports, and it's more than just anecdotal now, it was a very, very good night for Senator Hatch," his campaign spokesman Dave Hansen said, although no numbers were available.

Hatch was fighting to avoid the fate of his former Senate colleague, Utah's Bob Bennett, whose 2010 run for a fourth term foundered over conservative outrage at the healthcare overhaul pushed by President Barack Obama, leading to a revolt against the Republican establishment by state Tea Party activists.

Former state Senator Dan Liljenquist, 37, and state Representative Chris Herrod, 46, are among the most prominent of nine Republicans hoping to grab Hatch's traditional spot on the ballot, saying now is the time for change.

"It's time for a new generation to step up and lead," Liljenquist said in a recent interview. "Washington will not be changed from the inside."

Herrod described the caucuses as a "good night," adding it was hard to compete against Hatch's resources: "I think this is still a race the nation wants to watch."

Utah Republican Party Chairman Thomas Wright said preliminary estimates put turnout at between 125,000 to 200,000, more than double the number who took part in 2010.

The turnout was causing caucuses to run late and making it difficult for the party and candidates to get news of which delegates were being elected and who they supported.

Both of Hatch's challengers have gained a strong Tea Party following with campaigns focused on conservative themes such as smaller government and less tax. They have suggested Hatch's principles have been compromised by his lengthy tenure as part of the inside-the-Beltway establishment.

MAY NOT SEE CLEAR WINNER YET

Republican voters taking part in the hundreds of caucus meetings throughout Utah on Thursday in homes, churches, schools and other locations were selecting 4,000 delegates to represent them at a party convention on April 21.

The results will give a first significant sign of Hatch's re-election prospects, although a clear winner may not emerge until the convention, or later if a run-off vote is needed.

A candidate who emerges from the convention with 60 percent or more of the party's delegate vote will clinch the nomination.

If no contender takes 60 percent, the two highest vote getters compete in a primary election on June 26, the same day as Utah's presidential primary.

In 2010, Bennett came in third among delegates, and failed to make it out of the convention. Mike Lee won the primary runoff against the second-place Republican, and ultimately succeeded Bennett in the Senate.

Washington-based political action group FreedomWorks, closely aligned with the Tea Party movement, has spent nearly $600,000 to unseat Hatch this year, according to campaign expenditure filings.

But some longtime Republicans in Utah said the mood in the state was not the same as in 2010, when Bennett was pushed out, and that Hatch had positioned himself well going into the night.

"They underestimate how many people he has worked with over the years," said Enid Green Mickelson, a former congresswoman from Utah. "He has always worked hard for his re-election. He's always run like he's opposed."

To boost turnout, party officials stepped up precinct training and paid for lawn signs and television and radio ads reminding voters to attend.

"A real strong Tea Party member and good friend of mine, a little to the right of me, was bringing 50 people along who were supporting him to his precinct," said Swen Howard a legislative district vice-chair for the Republican Party in Davis County, just north of Salt Lake City.

The Mormon Church, which asked congregations to refrain from holding church meetings the same night as the party caucuses to avoid scheduling conflicts.

Ahead of the Bell: Unemployment Benefits

WASHINGTON (AP) ? The number of people seeking unemployment benefits likely declined last week, adding to evidence that the job market is strengthening.

Economists forecast that weekly applications dipped 4,000 last week to a seasonally adjusted 358,000, according to a survey by FactSet. The Labor Department will release the report at 8:30 a.m. Eastern time Thursday.

Applications have fallen 14 percent since October and last week were slightly above a four-year low. The four-week average, which smooths out fluctuations, remained roughly at 355,000. That's essentially unchanged from the previous week, which was also the lowest in four years.

The steady decline has coincided with the best three months of hiring in two years. From December through February, employers have added an average of 245,000 jobs per month. That's pushed down the unemployment rate to 8.3 percent, the lowest in three years. The rate fell for five straight months before holding steady in February.

When unemployment benefit applications drop consistently below 375,000, it usually signals that hiring is strong enough to lower the unemployment rate.

Better economic growth is spurring more job creation. The economy grew at an annual rate of 3 percent in the final three months of last year.

Other data suggest the recovery is gaining momentum. Consumers are more confident and have stepped up spending. Auto sales are rising. And the stock market keeps climbing ? the Dow Jones industrial average this week closed at its highest point since the last day of 2007.

The Federal Reserve is slightly more upbeat about the recovery, largely because of the surge in hiring. After its one-day meeting Tuesday, the Fed said unemployment should continue to decline gradually as the economy expands. And it noted that consumer spending and business investment have picked up.

The central bank took no further steps to aid the recovery and repeated its plan to keep short-term interest rates near zero through 2014.

One concern is that rising gas prices will force consumers to cut back on discretionary spending. That could weigh on economic growth and slow hiring. The Fed said it expects oil and gas prices to temporarily boost inflation but predicted longer-term inflation should remain stable.

Even with the improvement, the job market has a ways to go to fully recover from the Great Recession. More than 12.8 million people remain unemployed and the economy still has 5 million fewer jobs than before the downturn.

But the more robust job market has caused many so-called "discouraged workers" to start looking again. They're still unemployed, but they're back in the work force. The work force surged by 476,000 in February and almost 1 million the past two months.

If you've ever felt overwhelmed by deciding what brand of toothpaste to buy or what flight to book, two marketing professors think they know why.

"Decision quicksand," a painful element of 21st century life, ironically strikes hardest when people face trivial choices, say researchers Aner Sela of the University of Florida and Jonah Berger of the Wharton School, in a paper to be published later this year in the ?Journal of Consumer Research.?

While struggles to pick a new job or a select a mate might seem to demand the most deliberation, decision quicksand strikes even harder over trivial choices.? Little decisions cause a big problem precisely because they are surprisingly hard. Faced with too many options, consumers unconsciously connect difficulty with importance, and their brains are tricked into heavy deliberation mode, the researchers say in their paper, ?Decision Quicksand: How Trivial Choices Suck Us In."


?One could imagine a recursive loop between deliberation time, difficulty and perceived importance," write Sela and Berger.? "Inferences from difficulty may not only impact immediate deliberation but may kick off a cycle that leads people to spend more and more time on a decision that initially seemed rather unimportant."

The challenge of too many choices -- a bane of life in the age of information overload -- arises in part because people fail to recognize decisions as relatively unimportant.

"Why do we agonize over what toothbrush to buy, struggle with what sandwich to pick, and labor over which shade of white to paint the kitchen?? the authors ask. ?? Instead of realizing that picking a toothbrush is a trivial decision, we confuse the array of options and excess of information with decision importance, which then leads our brain to conclude that this decision is worth more time and attention."

But something else is going on, they contend: ?our brains are ruled by an unconscious force that mistakes difficulty ? any difficulty -- for importance.

To test their theory, the researchers set up numerous experiments. In one, volunteers were asked to select a flight using an online service.? Half the volunteers were forced to use a site with a small, hard-to-read font.? That one extra hurdle added nearly 50 percent to their deliberation time.? When told that the trip was short, so flight choice didn't matter as much, deliberation choice time spiked even more. (The researchers controlled for added time that could be blamed on simple difficulty reading.)

Decision struggles can be blamed for many poor outcomes ? couples? spats in the grocery store, or at the video rental place come to mind. But there are longer-term consequences.? Research shows that time spent in decision quicksand before a choice correlates with dissatisfaction after the fact.? And of course, there?s all that wasted time and emotional energy.

*Set decision rules and stick to them. In other words, start with a time limit that reflects the true importance of the choice. For example, "I will book a flight in 5 minutes, no matter what."

The Game Developers Conference is far from over ? actually, the show floor just opened today ? but if bumping into people over the past couple days has been anything to go by, a PSN game called Dyad seems to be getting the best word of mouth at the show. Well, let me try that again ? the Halo announcements, SimCity, Quantic Dream?s bundle of joy, and the Independent Games Festival probably win this award I just made up. But for the underdog word of mouth, I?m going with Dyad.

A couple days ago, I was able to play it for about an hour. Developer Shawn McGrath invited some media to try the game in his hotel room, each alone, with the lights off ? ?the cool kids? way of playing it,? he said ? which seemed odd, but made sense when I realized the game is a weird mash-up of Rez, a light show, a kaleidoscope, and a racing game. ?Weird? is his word, by the way.

"Jihad (holy war) is the only way to get the invading Americans out of Afghanistan," one banner read, before the protesters dispersed peacefully after about two hours.

The United States and its NATO allies are looking to withdraw their 130,000-odd combat troops from Afghanistan by 2014, although Sunday's massacre in Kandahar risks exacerbating a mood of war-weariness in the West.

In Washington, Obama warned the US public against a hasty drawdown from Afghanistan, after a weekend poll said most Americans believe the war is not worth the cost and want an early withdrawal.

"It's important for us to make sure that we get out in a responsible way, so that we don't end up having to go back in," Obama said in an interview with CBS station KDKA in Pittsburgh.

"But what we don't want to do, is to do it in a way that is just a rush for the exits," he said, stressing the need for an orderly withdrawal to get US personnel and equipment out, and to prevent Al-Qaeda rebuilding.

Yet while warning against a precipitous pullout, Obama said in a separate interview with Denver CBS affiliate KCNC that it was "important for us just to make sure that we are not... in Afghanistan longer than we need to be".

The Jalalabad students, echoing a call by the Afghan parliament on Monday, also demanded that the US soldier be tried in public in Afghanistan.

But the United States made clear that the soldier would be subject to US military law -- and that he could face execution if convicted.

Briefing reporters en route to a visit to Kyrgyzstan, Defense Secretary Leon Panetta was asked if the death penalty could apply. "My understanding is in these instances that could be a consideration," he said.

The US army sergeant was on his first tour of duty in Afghanistan after serving three tours in Iraq, according to US officials.

He left his forward operating base in Kandahar province in the pre-dawn darkness Sunday and went on a murderous rampage, before turning himself in back at the base, Afghan and American officials say.

He is accused of breaking into village homes and opening fire, killing 16 people including women and children, in an incident that has further imperilled Afghan-US relations as the countries try to craft a post-2014 partnership deal.

The soldier has not been named. But officials said he is in his 30s, and that the military investigation would look into whether he may have been suffering from mental trauma.

"War is hell. These kinds of events and incidents are going to take place," Panetta said, portraying the shooting as an isolated incident.

"We cannot allow these events to undermine our strategy or the mission that we're involved in. It's important that we push on."

Afghan President Hamid Karzai has described the shootings as "unforgivable", and the parliament declared that "people are running out of patience" over the behaviour of the foreign troops deployed in the country.

The Taliban, leading a 10-year insurgency against the foreign troops and Karzai's government, threatened to take revenge against "sick-minded American savages".

The massacre is the latest serious test of the US-Afghan alliance as the two countries pursue difficult talks on securing a strategic pact to govern their partnership once foreign combat troops leave Afghanistan in two years.

One major sticking point is the Karzai government's refusal so far to grant legal immunity to American troops -- the same issue that scuppered a US strategic pact with Iraq.

The massacre is also the latest in a series of actions by troops that has provoked outrage in Afghanistan, and comes weeks after the burning of the Korans sparked riots that killed 40 people and plunged ties to an all-time low.

'Chum Cam' Helps Catalog Endangered Sharks

Scientists have been trying to answer the question of whether marine protected areas, where sharks and their prey are off-limits to fishermen, are indeed home to more sharks than non-protected areas of the ocean.

But how to monitor an environment where humans can spend only a few hours at a time?

Researchers from New York's Stony Brook University solved this problem by setting up 200 baited underwater video cameras, nicknamed "chum cams," to count Caribbean reef sharks?(Carcharhinus perezi) inside and outside marine reserves on a reef in the Caribbean Sea.

Sharks, attracted by the smell of the bait, swam to the cameras, which allowed the research team to record, count and compare shark populations in two marine reserves to those in two areas where fishing is permitted.

Within the protected areas, sharks were caught on film at nearly four times as many camera sites as outside these areas, according to the research, which is published online today (March 8) in the journal PLoS One.

"Although we know that relatively sedentary reef fish and lobsters benefit from marine reserves, this study now presents visual proof that large, active sharks are also dramatically more abundant inside these protected areas, too," Mark Bond, lead author of the study and a doctoral student at Stony Brook University, said in a statement.

Caribbean reef sharks are listed as "near threatened" by the International Union for the Conservation of Nature, an independent body that assesses the conservation status of species around the world. Their dwindling numbers are due to intense fishing, according to the researchers.

Shark populations of many species around the world have declined precipitously in recent decades, many falling victim to the lucrative trade in fins for shark fin soup.

Scientists have been increasingly relying on cameras to count elusive and endangered terrestrial species, such as snow leopards.

"It is just as difficult to count sharks in the ocean, so we took a page from the big cat researchers' playbook and deployed baited video cameras to count the sharks," Demian Chapman, assistant professor at Stony Brook's School of Marine and Atmospheric Science, said in a statement.

"It's only fitting since these large apex predators are the 'big cats' of the sea," he said, "and like their feline counterparts, their continued existence on Earth is threatened."

Report: Twitter lost nearly $100 million in 2010 and early 2011, leaked finances show

Twitter lost nearly $100 million from the beginning of 2010 through early 2011 according to leaked financial information, the website Gawker reported Friday.

A "source with close knowledge of Twitter's financials" provided Gawker with information from 2010 and the first four months of 2011, the website said Friday. The popular San Francisco microblogging platform lost $67.8 million in the 2010 calendar year and $25.8 million from January through the end of April in 2011, according to figures provided to Gawker.

'American Idol' Report Card: Jessica Sanchez Owns The Night

Sanchez's take on Whitney Houston's 'I Will Always Love You' brings down the house as the guys tackle Stevie Wonder.

"American Idol" swung for the fences on Wednesday night — and we're not just talking about Steven Tyler's decision to wear his Roy Lichtenstein pants. The girls tackled Whitney Houston songs (borderline blasphemy?) and the guys did Stevie Wonder, and, after two hours of performances that ran the gamut from "startlingly great" to "shockingly unhinged" — with the majority falling somewhere in the middling middle — well, perhaps it's best if producers reined it in a tad.

Still, there were some highlights (Jessica Sanchez!), and with eliminations coming Thursday night (March 8) — though, in classic late-era "Idol," they've added some unnecessary wrinkles to the proceedings — here's our grades for the Top 13. Who passed with flying colors? Who flunked out in flaming fashion? Read on to find out!

Jessica Sanchez: Showed some major stones taking on the Whitney song to end all Whitney songs, "I Will Always Love You." Was maybe a touch too melismatic, didn't quite get there at the end, but still ... dang, girl. Tears and standing ovations both richly deserved. Grade: A

Hollie Cavanagh: Pint-sized powerhouse delivered the goods on "All the Man That I Need," including hitting that big, big note at the end. Showcased range, control and power — all that's missing is some personality. J.Lo sees her going all the way to the finals, and if Cavanagh keeps singing like this — and loses the nerves — she might be right. Grade: B+

Joshua Ledet: Ventured outside his comfort zone with a strutting, cutting take on Stevie's "I Wish," and took us all to church in the process. It was a bit screechy in parts, but he made up for it with amazing hand gestures and newfound confidence. Quoth Randy: "You wore it out!" We'd have to agree with that assessment. Grade: B+

Skylar Laine: The fact that she'd never sung Whitney is not nearly as shocking as her newfound dark-horse status. She took on "Where Do Broken Hearts Go," and though it started out a bit nasally (and sleepy), she picked up speed and ended up more or less killing it. "Biggest moment of the night," J.Lo said — of course, that was before Jessica Sanchez. Grade: B

Heejun Han: He proved to be equally adept at both humorous tweets and gross misspellings of celebrity names (seriously dude, it's "I-O-V-I-N-E"). His stripped-down take of "All in Love is Fair" was shaping up to be a slow-mo disaster, but he turned it around, and ended up putting his own silky smoove touch on the tune. We could do with less of his shtick, but the dude keeps delivering ... and flirting with J.Lo. Grade: B

Colton Dixon: Of course, was more than willing to take Jimmy and MJB's advice to perform one of his own songs, and, of course, it was super wussy. He ended up doing "Lately," which somehow managed to be both a tad flat and pitchy. Of course, only Randy seemed to notice. Girls seem to like his smoldering looks, which means he'll stick around. Grade: B-

Erika Van Pelt: Mary J called her tone "steak and potatoes," though her take on "I Believe In You and Me" was more Lean Cuisine. Spotty at the beginning, she pressed on, and ended up with a fairly legit — if slightly bland — performance. It gave J.Lo goosies, which makes you wonder if they had the air conditioner cranked up too high. She earned a nickname from Seacrest ("EVP"), so she seems safe. Grade: B-

Jeremy Rosado: Jer Bear put a sultry, supple spin on Stevie's "Ribbon in the Sky," and, in the process, made the most of his second chance. Sadly, Randy though it was lacking in "swag." Can we retire that phrase yet? Middling reviews from the judges didn't do him any favors. Grade: B-

Phillip Phillips: His night-closing "Pimp Spot" turned out to be not so pimp, since he had to follow Jessica Sanchez. From the minute they announced the guys were doing Stevie, I knew he was doing "Superstition," and lo and behold, he did just that. It was swampy, sorta scatty, and also sorta anti-climactic (and scary). Steven said he had a lot of "forget about it" in his voice ... don't worry, we already have. Grade: C

Deandre Brackensick: Skanked his way through "Master Blaster" and showed off his ill falsetto. It was sort of forgettable, yet also kind of fresh at the same time. We're glad Steven made the Naima comparison, since we didn't think we were allowed to go there. Irie! Grade: C

Jermaine Jones: The Gentle Giant's baritone was on full display with "Knocks Me Off My Feet." It started sort of bland, picked up steam toward the end, yet never truly seemed to connect. Quoth Steven: "That song fits you like an Armani suit" — that'd be a gigantic suit. Jones described his performance thusly: "I think I did a pretty good job." OK, dude. Grade: C-

Elise Testone: Ooof. Early attempts at "The Greatest Love of All" earned her the stank face from MJB, so (seemingly against her will) she ended up doing "I'm Your Baby Tonight." The end result was a near catatonic, downright uncomfortable performance. Randy said she was "Boxing with the song," and, well, she got knocked out. She seemed to come unglued during Seacrest's post-performance interview, and for someone who claimed "there's no excuse" for her spotty stint, she sure made a lot of excuses. Grade: D-

Shannon Magrane: Seacrest introduced her as "The pride of Tampa," though, after this debacle, he probably wants to reconsider that assessment. She did "I Have Nothing," which was a pretty apt metaphor for her entire performance. Sure, she's got a big voice, but she was swallowed up by the size and emotional wallop of the song itself — you know, because she's 16 or whatever. She tried to power her way through, but ended up just sort of shouting. If Erika Van Pelt is "EVP," she's "NVG" — not very good. Grade: F

Reversing Alzheimer's gene 'blockade' can restore memory, other cognitive functions

ScienceDaily (Feb. 29, 2012) ? MIT neuroscientists have shown that an enzyme overproduced in the brains of Alzheimer's patients creates a blockade that shuts off genes necessary to form new memories. Furthermore, by inhibiting that enzyme in mice, the researchers were able to reverse Alzheimer's symptoms.

The finding suggests that drugs targeting the enzyme, known as HDAC2, could be a promising new approach to treating the disease, which affects 5.4 million Americans. The number of Alzheimer's victims worldwide is expected to double every 20 years, and President Barack Obama recently set a target date of 2025 to find an effective treatment.

Li-Huei Tsai, leader of the research team, says that HDAC2 inhibitors could help achieve that goal, though it would likely take at least 10 years to develop and test such drugs.

"I would really strongly advocate for an active program to develop agents that can contain HDAC2 activity," says Tsai, director of the Picower Institute for Learning and Memory at MIT. "The disease is so devastating and affects so many people, so I would encourage more people to think about this."

Tsai and her colleagues report the findings in the Feb. 29 online edition of Nature. Lead author of the paper is Johannes Gr?ff, a postdoc at the Picower Institute.

Genome modification

Histone deacetylases (HDACs) are a family of 11 enzymes that control gene regulation by modifying histones -- proteins around which DNA is spooled, forming a structure called chromatin. When HDACs alter a histone through a process called deacetylation, chromatin becomes more tightly packaged, making genes in that region less likely to be expressed.

HDAC inhibitors can reverse this effect, opening up the DNA and allowing it to be transcribed.

In previous studies, Tsai had shown that HDAC2 is a key regulator of learning and memory. In the new study, her team discovered that inhibiting HDAC2 can reverse Alzheimer's symptoms in mice.

The researchers found that in mice with Alzheimer's symptoms, HDAC2 (but not other HDACs) is overly abundant in the hippocampus, where new memories are formed. HDAC2 was most commonly found clinging to genes involved in synaptic plasticity -- the brain's ability to strengthen and weaken connections between neurons in response to new information, which is critical to forming memories. In the affected mice, those genes also had much lower levels of acetylation and expression.

"It's not just one or two genes, it's a group of genes that work in concert to control different phases of memory formation," Tsai says. "With such a blockade, the brain really loses the ability to quickly respond to stimulation. You can imagine that this creates a huge problem in terms of learning and memory functions, and perhaps other cognitive functions."

The researchers then shut off HDAC2 in the hippocampi of mice with Alzheimer's symptoms, using a molecule called short hairpin RNA, which can be designed to bind to messenger RNA -- the molecule that carries genetic instructions from DNA to the rest of the cell.

With HDAC2 activity reduced, histone acetylation resumed, allowing genes required for synaptic plasticity and other learning and memory processes to be expressed. In treated mice, synaptic density was greatly increased and the mice regained normal cognitive function.

"This result really advocates for the notion that if there is any agent that can selectively down-regulate HDAC2, it's going to be very beneficial," Tsai says.

The researchers also analyzed postmortem brains of Alzheimer's patients and found elevated levels of HDAC2 in the hippocampus and entorhinal cortex, which play important roles in memory storage.

Reversing the blockade

The findings may explain why drugs that clear beta-amyloid proteins from the brains of Alzheimer's patients have offered only modest, if any, improvements in clinical trials, Tsai says.

Beta-amyloid proteins are known to clump in the brains of Alzheimer's patients, interfering with a type of cell receptor needed for synaptic plasticity. The new study shows that beta amyloid also stimulates production of HDAC2, possibly initiating the blockade of learning and memory genes.

"We think that once this epigenetic blockade of gene expression is in place, clearing beta amyloid may not be sufficient to restore the active configuration of the chromatin," Tsai says.

The appeal of HDAC2 inhibitors, Tsai says, is that they could conceivably reverse symptoms even after the blockade is well-established. However, much more drug development has to take place before such a compound could enter clinical trials. "It's really hard to predict," Tsai says. "Clinical trials would probably be five years down the line. And if everything goes well, to become an approved drug would probably take at least 10 years."

Some general HDAC inhibitors, not specific to HDAC2, have been tested in clinical trials as cancer drugs. However, to treat Alzheimer's, a more selective approach is needed, Tsai says. "You want something as selective as possible, and as safe as possible," she says.
